生化和生理学机理 | The U1 small molecule nuclear ribonucleoprotein (U1 snRNP) antigen is an RNP/Sm multi enzyme complex composed of three U1 specific ribonucleoproteins (RNP-A, RNP-C, RNP-68K) and different Sm nucleoproteins (B/B ', D, E, F, G). RNP/Sm complex is an extractable nuclear antigen (ENA) that targets self reactive B and T cells in various rheumatic diseases. Anti RNP antibodies bind to peptides specific for U1 snRNA (RNP-A, RNP-C, RNP-68K), while anti Sm antibodies can bind to peptides specific for U1, U2, U5, and U4/6 snRNA. High titer anti-U1 snRNP autoantibodies are one of the important criteria for diagnosing mixed connective tissue diseases (MCTD, Sharpe syndrome), with a positivity rate of up to 95% -100%, and antibody titers are correlated with disease activity. This antibody can also be detected in 30% -40% of systemic lupus erythematosus (SLE) patients, but almost always accompanied by anti Sm antibodies. Anti-U1-snRNP autoantibodies can also be found in the serum of patients with other rheumatic and immune diseases, including dermatomyositis (10% -20%), unclassified connective tissue disease (20%), Sjogren's syndrome (20%), scleroderma (7%), and overlap syndrome (27%). |
